Diadaptasi dari dongeng karya E.B. White's berjudul sama, "Charlotte's Web", mengangkat kisah tentang persahabatan hewan dan manusia. Fern (Dakota Fanning) adalah satu-satunya anak yang percaya kalau Wilbur adalah seekor babi spesial. Itulah sebabnya Fern mencegah agar Wilbur tak disembelih seperti babi-babi di peternakannya.
Menjalani kehidupannya, Wilbur yang pemalu bersahabat dengan seekor laba-laba bernama Charlotte A. Cavatic (Julia Roberts). Keakraban yang terjalin itu mengilhami binatang lainnya untuk berkumpul sebagai satu keluarga. Ketika tiba waktunya bagi Wilbur untuk disembelih, Charlotte dengan ide kreatifnya menciptakan keajaiban yang membuat Wilbur bukan hanya sekedar babi biasa.
